development of new protocols for plant regeneration from protoplasts and organ tissues, induction of callus and suspension culture in vitro, isolation and cultivation of protoplasts, microclonal propagation, protoplast fusion, mutagenesis and selection procedures.

Agrobacterium-mediated (including floral dip method), direct gene transfer by microprojectile bombardment, electroporation of protoplasts, polyethylene glycol-mediated, etc.

cDNA library screening, gene cloning, construction of all types of vectors for transient and stable (trans)gene expression in bacterial, plant, and animal systems, analyses of spatiotemporal promoter activity in vivo using GFP and GUS reporter gene system, identification and deletion analyses of regulatory elements in promoters, isolation and Southern analyses of genomic, chloroplast and mitochondrial DNA, design and synthesis of PCR primers, conventional PCR, RACE-PCR, analyses of gene expression (northern blotting, differential display, RT-PCR, etc.) and protein accumulation (AU-PAGE, western blotting and immunodetection, histochemical localization), detection of enzyme activity (e.g. fluorometric GUS assay), isoenzyme analyses.
